چکیده

UNLABELLED PREMISE OF THE STUDY Microsatellite primers were developed in the hemiparasitic perennial forb Castilleja sessiliflora to investigate patterns of gene flow and genetic diversity within and among populations. • METHODS AND RESULTS Twelve polymorphic loci were identified in C. sessiliflora and tested on three populations (32 individuals each) sampled across the range of the species. The loci amplified di- and trinucleotide repeats with 3-14 alleles per locus. To assess cross-amplification, primer pairs were also tested on 24 additional Castilleja species that represent the morphological and geographic diversity of the genus. We provide reports of their effectiveness in all 25 taxa. • CONCLUSIONS These results indicate the utility of these primers in C. sessiliflora for future studies of genetic structure and gene flow, as well as their widespread applicability in other members of the diverse and complex genus Castilleja.

Primers for Castilleja and their utility across Orobanchaceae: II. Single-copy nuclear loci1

PREMISE OF THE STUDY We developed primers targeting nuclear loci in Castilleja with the goal of reconstructing the evolutionary history of this challenging clade. These primers were tested across other major clades in Orobanchaceae to assess their broader utility. METHODS AND RESULTS We assembled low-coverage genomes for three taxa in Castilleja and developed primer combinations for the singl...

Primers for Castilleja and their utility across Orobanchaceae: I. Chloroplast primers1

PREMISE OF THE STUDY Chloroplast primers were developed from genomic data for the taxonomically challenging genus Castilleja. We further tested the broader utility of these primers across Orobanchaceae, identifying a core set of chloroplast primers amplifying across the clade. METHODS AND RESULTS Using a combination of three low-coverage Castilleja genomes and sequence data from 12 Castilleja...

Homoploid hybrid speciation in a rare endemic Castilleja from Idaho (Castilleja christii, Orobanchaceae).

UNLABELLED PREMISE OF THE STUDY Hybridization is an important evolutionary force in the history of angiosperms; however, there are few examples of stabilized species derived through homoploid hybrid speciation. Homoploid hybrid species are generally detected via the presence of genetic additivity of parental markers, novel ecological and spatial distinctions, and novel morphological traits, ...

Tangled trios?: Characterizing a hybrid zone in Castilleja (Orobanchaceae).

Hybridization and polyploidization are exceedingly important processes because both influence the ecological envelope and evolutionary trajectory of land plants. These processes are frequently invoked for Castilleja (Indian paintbrushes) as contributors to morphological and genetic novelty and as complicating factors in species delimitations. Here, we provide a detailed analysis of morphologica...

The evolutionary origin of a second radiation of annual Castilleja (Orobanchaceae) species in South America: The role of long distance dispersal and allopolyploidy.

Considerable attention has been directed toward understanding the wide gaps in range that are common among many groups of closely related organisms. By placing their biology and geography in a phylogenetic context, we may gain a broader knowledge of the series of historical events that have led to present species distributions. In addition to the North American annuals, a second radiation of an...
